Ketotifen therapy, a mast cell blocker, significantly reduced pain and slowed tumor growth in neurofibromatosis patients. This treatment also improved patients

    Area of Science:

    • Oncology
    • Dermatology
    • Pharmacology

    Background:

    • Neurofibromas are tumors associated with neurofibromatosis.
    • Mast cells are abundant in neurofibromas and may promote tumor growth.
    • Ketotifen is a mast cell stabilizer that blocks mast cell secretion.

    Observation:

    • A patient with severe neurofibromatosis (NF) was treated with ketotifen.
    • Eleven patients with symptomatic neurofibromas received ketotifen (2-4 mg/d) for 30-43 months.
    • Total treatment duration was 389 patient-months (32.4 patient-years).

    Findings:

    • All patients experienced reduced neurofibroma-associated pruritus, pain, and tenderness.
    • A consistent decrease in the rate of neurofibroma growth was observed.
    • Patients reported improved well-being, productivity, and general performance.

    Related Experiment Videos

    Implications:

    • Mast cell secretions likely contribute to neurofibroma growth and symptoms.
    • Mast cell blockers, such as ketotifen, may effectively retard neurofibroma growth.
    • Ketotifen therapy shows potential for managing neurofibromatosis symptoms and progression.
