Nesfatin-1 administration, both centrally and peripherally, significantly increases respiratory rate and ventilation in rats. These hyperventilatory effects are influenced by food intake and administration method.
Area of Science:
- Neuroscience
- Physiology
- Endocrinology
Background:
- Nesfatin-1 is an anorexiogenic peptide involved in central nervous system functions like cardiovascular control and energy balance.
- It is expressed in brain nuclei regulating respiration, but its direct respiratory effects were unstudied.
- This peptide acts as a neurotransmitter and/or neuromodulator.
Purpose of the Study:
- To investigate the impact of central and peripheral nesfatin-1 administration on respiratory parameters in rats.
- To compare these effects in fasted versus fed states.
- To explore dose- and time-dependency of nesfatin-1's respiratory actions.
Main Methods:
- Rats were administered nesfatin-1 via intracerebroventricular (ICV) or intravenous (IV) injection.
- Respiratory parameters including tidal volume (TV), respiratory rate (RR), and respiratory minute ventilation (RMV) were measured.
- Experiments were conducted on both 12h-fasted and fed-ad libitum rats.
Main Results:
- ICV nesfatin-1 induced significant hyperventilation (increased TV, RR, RMV) in both fasted and fed rats, in a dose- and time-dependent manner.
- Central nesfatin-1's hyperventilatory effects were more pronounced in fed rats.
- IV nesfatin-1 increased RR and RMV but not TV in fed rats.
Conclusions:
- Both central and peripheral nesfatin-1 administration elicit significant hyperventilatory responses in rats.
- The magnitude of these respiratory effects is dependent on the rat's feeding status and the route of nesfatin-1 delivery.
- Nesfatin-1 represents a novel factor influencing respiratory control.
