High-dose methotrexate in malignant melanoma.

C P Karakousis, M Carlson

    Cancer Treatment Reports
    |August 1, 1979
    PubMed
    Summary

High-dose methotrexate showed limited efficacy in advanced malignant melanoma patients resistant to prior treatments. While one patient achieved an objective response, seven experienced disease stabilization, with notable toxicity observed.

    Area of Science:

    • Oncology
    • Pharmacology

    Background:

    • Advanced malignant melanoma presents a significant therapeutic challenge.
    • Patients often develop resistance to conventional chemotherapy protocols.

    Purpose of the Study:

    • To evaluate the efficacy and toxicity of high-dose methotrexate in patients with advanced malignant melanoma.
    • To assess treatment outcomes in a heavily pre-treated patient population.

    Main Methods:

    • A cohort of 28 patients with advanced malignant melanoma was treated with high-dose methotrexate.
    • Patients had previously failed at least two chemotherapy regimens.

    Main Results:

    • One patient (approximately 3.6%) achieved an objective response.
    • Seven patients (25%) experienced disease stabilization, with an average duration of 4 months.
    • Five instances of serious, non-fatal toxicity were reported.

    Conclusions:

    • High-dose methotrexate demonstrates minimal objective response rates in advanced, pre-treated malignant melanoma.
    • Disease stabilization is achievable in a subset of patients, but toxicity must be considered.
