Summary

Insulin-like growth factor 2 (IGF2) and insulin-like growth factor 1 receptor (IGF1R) mRNAs are present in human sperm. Higher levels of these mRNAs correlate with increased sperm concentration and total sperm count.

Detection of Axonally Localized mRNAs in Brain Sections Using High-Resolution In Situ Hybridization

RNA in situ hybridization (ISH) enables the visualization of RNAs in cells and tissues. Here we show how combination of RNAscope ISH with immunohistochemistry or histological dyes can be successfully used to detect mRNAs localized to axons in sections of mouse and human...

Phosphopeptide Analysis of Rodent Epididymal Spermatozoa

Proteomic analysis of any cell type is highly dependent on both purity and pre-fractionation of the starting material in order to de-complexify the sample prior to liquid chromatography mass spectrometry (MS). By using back-flushing techniques, pure spermatozoa can be obtained from rodents. Following digestion, phosphopeptides can be enriched using...

A Technique for Percutaneous Epididymal Spermatozoa Aspiration and Processing

Source:Li Xiaoming1, Liu Xiao1, Li Haiyang1, Cai Xiaohua1, Ou Jianping11Center for Reproductive Medicine, The Third Affiliated Hospital of Sun Yat-sen University. This video demonstrates the percutaneous aspiration of epididymal spermatozoa from a patient with azoospermia, a condition characterized by the absence of spermatozoa in the semen. The procedure involves disinfecting the scrotal area, aspirating epididymal fluid using a syringe with a specialized culture solution and processing the...

High-Resolution Respirometry to Assess Mitochondrial Function in Human Spermatozoa

The analysis of sperm mitochondrial function by high-resolution respirometry permits the measurement of the oxygen consumption of freely moving spermatozoa in a closed-chamber system. The technique can be applied to measure respiration in human spermatozoa, which provides information on sperm mitochondrial characteristics and...

Visualization of Endoplasmic Reticulum Localized mRNAs in Mammalian Cells

Here we describe a method to visualize endoplasmic reticulum-associated mRNAs in mammalian tissue culture cells. This technique involves the selective permeabilization of the plasma membrane with digitonin to remove cytoplasmic contents followed by fluorescent in situ hybridization to detect either bulk poly(A) mRNA or specific...
