Summary

Blood DNA methylation profiles can predict the future course of major depressive disorder (MDD). This epigenetic signature may help identify individuals at risk for chronic or recurrent MDD, aiding clinical decisions.

Long-term Depression

Long-term depression, or LTD, is one of the ways by which synaptic plasticity—changes in the strength of chemical synapses—can occur in the brain. LTD is the process of synaptic weakening that occurs over time between pre and postsynaptic neuronal connections. The synaptic weakening of LTD works in opposition to synaptic strengthening by long-term potentiation (LTP) and together are the main mechanisms that underlie learning and memory.

Design and Implementation of an fMRI Study Examining Thought Suppression in Young Women with, and At-risk, for Depression

We aim to identify the neural correlates underlying sustained and transient thought suppression, and thought re-emergence in controls, at-risk and depressed individuals. Activation was greatest for controls compared to the at-risk and the depressed group in the dorsolateral prefrontal cortex during thought suppression and anterior cingulate cortex during thought...

In vitro Methylation Assay to Study Protein Arginine Methylation

Protein arginine methylation, catalyzed by a class of enzymes viz., protein arginine methyl transferases (PRMTs), is the process of enzymatic addition of methyl group(s) to arginines within proteins. The in vitro methylation assay is the most dependable tool for assessing the methylation status of known or novel PRMT...

Optogenetic Induction of Long-Term Depression for Suppressing Narcotic-Seeking Behavior in Rats

Source: Rich, M. T., et al. Using Optogenetics to Reverse Neuroplasticity and Inhibit Cocaine Seeking in Rats. J. Vis. Exp. (2021)This video demonstrates the use of optogenetics to induce long-term depression (LTD) at medial geniculate nucleus to lateral amygdala (MGN-LA) synapses, weakening cue-induced narcotic-seeking behavior in a rat model. The rat, surgically implanted with optical fibers and virally transduced to express light-sensitive ion channels, undergoes optogenetic stimulation with...

Assessment of Long-term Depression Induction in Adult Cerebellar Slices

In some gene-manipulated animals, using a single protocol may fail to induce LTD in cerebellar Purkinje cells, and there may be a discrepancy between LTD and motor learning. Multiple protocols are necessary to assess LTD-induction in gene-manipulated animals. Standard protocols are...
