Optogenetic Entrainment of Hippocampal Theta Oscillations in Behaving Mice07:33

Optogenetic Entrainment of Hippocampal Theta Oscillations in Behaving Mice

12.2K
We describe the use of optogenetics and electrophysiological recordings for selective manipulations of hippocampal theta oscillations (5-10 Hz) in behaving mice. The efficacy of the rhythm entrainment is monitored using local field potential. A combination of opto- and pharmacogenetic inhibition addresses the efferent readout of hippocampal...

10.3K
Here, we present a protocol for recording rhythmic neuronal network theta and gamma oscillations from an isolated whole hippocampal preparation. We describe the experimental steps from extraction of the hippocampus to details of field, unitary and whole-cell patch clamp recordings as well as optogenetic pacing of the theta...

This video demonstrates a protocol to modulate theta oscillations in a genetically engineered mouse model with light-sensitive ion channels in its GABAergic medial septum neurons. By delivering light pulses to the hippocampus, the protocol aims to synchronize neuronal activity and generate theta oscillations, which are recorded as local field...

Automatic Detection of Highly Organized Theta Oscillations in the Murine EEG

9.7K
Theta activity in the hippocampus is related to specific cognitive and behavioral stages. Here, we describe an analytical method to detect highly-organized theta oscillations within the hippocampus using a time-frequency (i.e., wavelet analysis)-based...

This video demonstrates the recording of rhythmic neuronal network activity, also known as theta oscillations, in an isolated hippocampus. Electrodes are inserted into various layers and spatial positions to detect and analyze theta oscillations. This shows the unique neuronal compositions and the connectivity of the neuronal network across different layers of the...

Assessment of Social Transmission of Food Preferences Behaviors

8.4K
This paper presents a protocol for the investigation of social transmission of food preference in mice. The advantages and possible applications for this procedure, for instance, in detecting early changes in AD mouse models, are highlighted. To conclude, interpretation of the results in light of critical details are...

Summary

Synchronized brain waves between the ventral hippocampus (vHPC) and medial prefrontal cortex (mPFC) are crucial for avoidance behavior. Optogenetic stimulation at 8 Hz enhanced this synchrony, increasing avoidance.

Area of Science:

  • Neuroscience
  • Behavioral Neuroscience
  • Neural Oscillations

Background:

  • Long-range synchronization of neural oscillations is linked to behaviors, but its causal role is not established.
  • Avoidance behavior in mice shows increased theta-frequency (∼8 Hz) synchrony between the ventral hippocampus (vHPC) and medial prefrontal cortex (mPFC).

Purpose of the Study:

  • To investigate the causal role of vHPC-mPFC synchrony in avoidance behavior.
  • To determine if optogenetic modulation of vHPC-mPFC activity influences avoidance behavior.

Main Methods:

  • Used optogenetics to dynamically modulate vHPC-mPFC terminal activity in mice.
  • Applied oscillatory and pulsatile stimulation at various frequencies (2, 4, 8, 20 Hz).
  • Measured avoidance behavior and assessed vHPC-mPFC neurotransmission and neural entrainment.

Main Results:

  • 8 Hz oscillatory stimulation maximally increased avoidance behavior compared to other frequencies and pulsatile stimulation.
  • Stimulation enhanced vHPC-mPFC neurotransmission and entrained neural activity.
  • Increased synchrony was observed between vHPC theta activity and mPFC spiking.

Conclusions:

  • vHPC-mPFC theta-frequency communication plays a critical role in generating avoidance behavior.
  • Synchronized neural oscillations causally facilitate neural transmission and behavior.
