Summary

Mechanical loading, like weight-bearing activities, is crucial for preventing bone loss in arthritis. Blocking mechanical loading worsened bone loss, while enhancing it helped preserve bone mass.

Area of Science:

  • Orthopedics
  • Rheumatology
  • Bone Biology

Background:

  • Arthritis causes bone loss through inflammation and reduced physical activity.
  • Mechanical loading is vital for maintaining bone mass; disuse leads to bone loss.
  • The role of mechanical loading in arthritis-related bone changes is not fully understood.

Purpose of the Study:

  • To investigate the impact of mechanical loading on periarticular bone and inflammation during arthritis.
  • To determine if blocking or enhancing mechanical loading affects arthritis-induced bone loss and inflammation.

Main Methods:

  • Arthritis was induced and mechanical loading was manipulated using botulinum neurotoxin A (Botox) or cyclic compressive loading.
  • Arthritis severity was assessed histologically.
  • Bone mass (trabecular and cortical) was measured using micro-computed tomography (μCT).
  • Subchondral osteoclastogenesis and bone turnover markers were analyzed.

Main Results:

  • Inhibition of mechanical loading exacerbated arthritis-induced bone loss but did not alter inflammation.
  • Enhanced mechanical loading mitigated arthritis-induced bone loss.
  • Mechanical loading partially blocked arthritis-induced increases in bone resorption markers.

Conclusions:

  • Muscle forces and mechanical loading play a significant role in preventing bone loss during arthritis.
  • Weight-bearing activities may counteract arthritis-mediated bone loss.
