Summary

Chronic hypercholesterolemia in apolipoprotein E-deficient mice impairs retinal arteriole function and increases oxidative stress. However, antioxidant defenses prevent neuron and axon loss in the retina.

Background:

  • Atherogenic lipoproteins can cause tissue damage through impaired perfusion and oxidative stress.
  • Endothelial dysfunction and cell loss in the retina are potential consequences of chronic hypercholesterolemia.

Purpose of the Study:

  • To test the hypothesis that chronic hypercholesterolemia induces endothelial dysfunction and cell loss in the retina.
  • To investigate the role of apolipoprotein E deficiency in retinal vascular and neuronal health.

Main Methods:

  • Apolipoprotein E-deficient (ApoE-/-) mice and wild-type controls were used.
  • Retinal arteriole responses, oxidative and reactive nitrogen species, LOX-1 and NADPH oxidase expression, and redox gene expression were analyzed.
  • Retinal ganglion cell layer cell number and optic nerve axon number were quantified.

Main Results:

  • ApoE-/- mice exhibited impaired retinal arteriole responses to acetylcholine.
  • Increased expression of LOX-1 and NOX2, and elevated reactive oxygen species were observed in retinal blood vessels.
  • Despite increased oxidative stress markers and altered gene expression, retinal cell and axon numbers remained unchanged.

Conclusions:

  • Apolipoprotein E deficiency leads to endothelial dysfunction and oxidative stress in retinal arterioles, potentially causing retinal hypoxia.
  • Upregulation of antioxidant enzymes protects nonvascular retinal tissue from oxidative stress, preserving neurons and axons.
