Summary

RADA16-I hydrogel shows promise for regenerating recurrent laryngeal nerves (RLNs) in rats. This study found improved nerve regeneration and muscle preservation, suggesting potential for clinical application in RLN repair.

Background:

  • Recurrent laryngeal nerve (RLN) injuries often result in insufficient functional recovery.
  • Existing treatments for RLN regeneration lack consistent effectiveness.
  • RADA16-I, a self-assembling peptide, promotes neurite outgrowth and synapse formation in vitro.

Purpose of the Study:

  • To investigate the efficacy of RADA16-I hydrogels in promoting the regeneration of transected recurrent laryngeal nerves in a rat model.

Main Methods:

  • Fifteen rats underwent transection of the recurrent laryngeal nerve, with gaps bridged by silicone tubes.
  • The RADA16-I (+) group received RADA16-I hydrogel injection into the tube.
  • Control groups included RADA16-I (-) and neurectomy.
  • Evaluations at 8 weeks included laryngoscopy, electrophysiology, and histology.

Main Results:

  • Partial vocal fold movement recovery was observed in one rat treated with RADA16-I hydrogel.
  • Significantly higher neurofilament-positive areas and myelinated nerve counts were found in the RADA16-I (+) group compared to the RADA16-I (-) group.
  • The thyroarytenoid muscle area was significantly larger in the RADA16-I (+) group than in the neurectomy group.

Conclusions:

  • RADA16-I hydrogel demonstrated effectiveness in promoting recurrent laryngeal nerve regeneration.
  • The findings suggest RADA16-I hydrogel as a potential therapeutic agent for RLN repair.
