Summary

Major Depressive Disorder (MDD) is linked to impaired social functioning. Diminished Default Mode Network (DMN) connectivity in the brain may underlie this social dysfunction in MDD patients.

Purpose of the Study:

  • To investigate the relationship between Default Mode Network (DMN) intrinsic functional connectivity and social dysfunction in patients with Major Depressive Disorder (MDD).
  • To explore whether DMN connectional integrity is associated with social deficits in MDD.

Main Methods:

  • Resting-state functional magnetic resonance imaging (fMRI) data from 74 MDD patients were analyzed using Independent Component Analysis and Dual Regression.
  • Categorical and dimensional analyses examined DMN connectivity in relation to social dysfunction (loneliness, social disability, small social network).
  • Statistical thresholding and multiple comparisons correction were performed using Threshold-Free Cluster Enhancement (TFCE) with family-wise error (FWE) correction (P < 0.05).

Main Results:

  • Greater social dysfunction in MDD patients was cautiously linked to diminished DMN connectivity.
  • Specific regions showing reduced connectivity included the rostromedial prefrontal cortex and posterior superior frontal gyrus.
  • These findings suggest a potential neurobiological basis for social deficits in MDD.

Conclusions:

  • Alterations in DMN connectivity may be relevant to social dysfunction observed in Major Depressive Disorder.
  • This study provides preliminary evidence linking DMN connectional integrity to social functioning in MDD.
  • Further research is warranted to elucidate the precise role of DMN in MDD-related social impairments.
