Background:
MicroRNAs (miRNAs) are endogenous, single-stranded, non-coding RNAs acting as negative regulators of gene expression involved in a number of physiological processes. MiRNAs' expression is commonly dysregulated in many types of human tumor diseases and cancers, including thyroid cancers, and is often involved in tumor initiation and progression. miR-19a, a member of miR-17-92 cluster, has been demonstrated to promote cell growth in anaplastic thyroid cancer (ATC), the most advanced and aggressive thyroid cancer.
Purpose:
In this work, we investigate the potential contribution of miR-19a in thyroid cancer cells poor prognosis and de-differentiation.
Methods:
We directly modulated the expression of miR-19a in papillary (PTC) and anaplastic thyroid carcinoma cell lines through transfection of specific miR-19a mimic or inhibitor. Further, we performed gene expression analysis of specific genes to evaluate miR-19a association with cell cycle, differentiation, and poor prognosis.
Results:
Our data indicate that miR-19a overexpression in PTC cells significantly promotes cell growth, decreases the expression of differentiation genes and activates poor prognosis genes. Its inhibition in ATC cells reduces cell proliferation and the expression of genes related to poor prognosis but does not affect differentiation.
Conclusion:
Our findings reveal the existence of functional associations between miR-19a expression and thyroid cancer progression and malignancy suggesting miR-19a as a novel candidate therapeutic target for ATC.
Insights
MicroRNA-19a (miR-19a) promotes thyroid cancer growth and malignancy. Inhibiting miR-19a in aggressive anaplastic thyroid cancer (ATC) reduces proliferation, suggesting miR-19a as a therapeutic target.
Area of Science:
- Molecular Biology
- Genetics
- Oncology
Background:
- MicroRNAs (miRNAs) are key regulators of gene expression, often dysregulated in cancers.
- miR-19a, part of the miR-17-92 cluster, is implicated in promoting anaplastic thyroid cancer (ATC) cell growth.
Purpose of the Study:
- To investigate the role of miR-19a in thyroid cancer cell de-differentiation and poor prognosis.
- To explore miR-19a's contribution to the progression of papillary (PTC) and anaplastic thyroid carcinoma (ATC).
Main Methods:
- Modulation of miR-19a expression in PTC and ATC cell lines using mimics and inhibitors.
- Gene expression analysis to assess miR-19a's impact on cell cycle, differentiation, and prognosis-related genes.
Main Results:
- Overexpression of miR-19a in PTC cells enhanced cell growth, reduced differentiation markers, and activated poor prognosis genes.
- Inhibition of miR-19a in ATC cells decreased proliferation and expression of poor prognosis genes, without affecting differentiation.
Conclusions:
- miR-19a expression is functionally linked to thyroid cancer progression and malignancy.
- miR-19a represents a potential therapeutic target for advanced thyroid cancers like ATC.

Tumor Progression
Colon cancer is one of the best-documented examples of tumor progression. Early mutation in the APC gene in colon cells causes a small growth on the colon wall called a polyp. With time, this polyp grows into a benign, pre-cancerous tumor. Further...
