Summary

Elongator complex subunit Elp3 is crucial for zebrafish trunk development. Its knockdown reduces tRNA modification mcm5s2U and sonic hedgehog (Shh) signaling, which can be rescued by Shh activation.

Area of Science:

  • Molecular Biology
  • Developmental Biology
  • RNA Biology

Background:

  • Transfer RNAs (tRNAs) undergo extensive post-transcriptional modifications.
  • Modifications in the tRNA anti-codon loop are vital for accurate decoding.
  • The Elongator complex synthesizes the mcm5s2U modification, essential for tRNA function.

Purpose of the Study:

  • To investigate the role of Elp3, the catalytic subunit of the Elongator complex, in zebrafish development.
  • To elucidate the molecular mechanisms linking Elp3 function to developmental processes.

Main Methods:

  • Zebrafish model system utilized for developmental studies.
  • Knockdown of the elp3 gene to assess its developmental impact.
  • Analysis of mcm5s2U levels and sonic hedgehog (Shh) signaling activity.
  • Functional rescue experiments by activating the Shh pathway.

Main Results:

  • elp3 knockdown in zebrafish resulted in impaired trunk development.
  • Reduced levels of the tRNA modification mcm5s2U were observed in elp3 knockdown animals.
  • Sonic hedgehog (Shh) signaling activity was diminished following elp3 knockdown.
  • Activation of the Shh pathway successfully reverted the trunk development defects.

Conclusions:

  • Elp3 is essential for normal trunk development in zebrafish.
  • A functional link exists between Elongator complex activity, tRNA modification (mcm5s2U), and Shh signaling.
  • The findings suggest a novel molecular mechanism involving Elongator and Shh in development.
