Summary
MicroRNAs (miRNAs) in peripheral blood mononuclear cells (PBMCs) show altered expression in breast and colon cancers. This panel of miRNAs, particularly miR-21, may serve as novel biomarkers for early cancer detection.
Area of Science:
- Oncology
- Molecular Biology
- Biomarker Discovery
Background:
- Aberrant microRNA (miRNA) expression is implicated in cancer development and progression.
- miRNAs are being investigated as potential diagnostic, prognostic, and monitoring tools in oncology.
- Specific miRNAs (miR-21, miR-31, miR-143, miR-145) are differentially expressed in cancer patients.
Purpose of the Study:
- To characterize the expression of miR-21, miR-31, miR-143, and miR-145 in peripheral blood mononuclear cells (PBMCs) of breast and colorectal cancer patients.
- To compare miRNA expression in cancer patients versus healthy controls.
- To evaluate the potential of these miRNAs as tumor markers for breast and colon cancer.
Main Methods:
- Study included 30 breast cancer patients, 30 colorectal cancer patients, and 30 healthy controls.
- Peripheral blood mononuclear cells (PBMCs) were isolated from blood samples.
- miRNA expression was analyzed using a two-step reverse transcription and quantitative polymerase chain reaction (qPCR) method.
Main Results:
- In breast cancer, miR-143 and miR-31 decreased, while miR-145 and miR-21 increased compared to controls.
- In colon cancer, miR-143, miR-145, and miR-21 increased, with no significant change in miR-31.
- miR-21 showed the highest increase in both cancer types; miR-143 correlated with colon cancer TNM stage.
Conclusions:
- A biomarker panel of miR-21, miR-31, miR-143, and miR-145 in PBMCs could offer a new diagnostic approach for early breast and colon cancer detection.
- miR-21, with its elevated expression, is a promising tumor biomarker and potential therapeutic target for breast and colon cancers.

MicroRNAs

MicroRNA (miRNA) are short, regulatory RNA transcribed from introns (non-coding regions of a gene) or intergenic regions (stretches of DNA present between genes). Several processing steps are required to form biologically active, mature miRNA. The initial transcript, called primary miRNA (pri-mRNA), base-pairs with itself, forming a stem-loop structure. Within the nucleus, an endonuclease enzyme, called Drosha, shortens the stem-loop structure into hairpin-shaped pre-miRNA. After the pre-miRNA...

In humans, more than 80% of the genome gets transcribed. However, only around 2% of the genome codes for proteins. The remaining part produces non-coding RNAs which includes ribosomal RNAs, transfer RNAs, telomerase RNAs, and regulatory RNAs, among other types. A large number of regulatory non-coding RNAs have been classified into two groups depending upon their length – small non-coding RNAs, such as microRNA, which are less than 200 nucleotides in length, and long non-coding RNA...
