Growth differentiation factor 15 (GDF15) knockdown enhances ferroptosis in MGC803 cancer cells. This occurs by reducing solute carrier family 7 member 11 (SLC7A11) expression and system Xc- function, promoting cell death.
Area of Science:
- Cell Biology
- Cancer Research
- Biochemistry
Background:
- Ferroptosis is an iron-dependent regulated cell death pathway implicated in cancer.
- Growth differentiation factor 15 (GDF15) influences cancer cell properties, but its role in ferroptosis is unknown.
- Solute carrier family 7 member 11 (SLC7A11) is a key regulator of ferroptosis via the system Xc- transporter.
Purpose of the Study:
- To investigate the role of GDF15 in ferroptosis.
- To determine the effect of GDF15 on SLC7A11 expression and system Xc- function.
- To elucidate the mechanism by which GDF15 influences ferroptosis in MGC803 cells.
Main Methods:
- GDF15 knockdown in MGC803 cells.
- CCK8 assay to assess cell viability and ferroptosis.
- Quantitative real-time PCR (qRT-PCR) and Western blotting to measure gene and protein expression.
- Measurement of extracellular glutamate, intracellular glutathione (GSH), and lipid reactive oxygen species (ROS).
Main Results:
- GDF15 knockdown decreased SLC7A11 expression.
- GDF15 knockdown promoted erastin-induced ferroptosis in MGC803 cells.
- GDF15 inhibition attenuated erastin-induced SLC7A11 upregulation.
- GDF15 knockdown led to decreased extracellular glutamate and intracellular GSH, and increased lipid ROS upon erastin treatment.
Conclusions:
- GDF15 knockdown promotes erastin-induced ferroptosis in MGC803 cells.
- This promotion is mediated by the attenuation of SLC7A11 expression and system Xc- function.
- GDF15 plays a significant role in regulating ferroptosis, potentially through modulation of the SLC7A11/system Xc- axis.
