Mouse Models of Cancer Study02:43

Mouse Models of Cancer Study

6.3K
Mice have long served as models for studying human biology and pathology because of their phylogenetic and physiological similarity with humans. They are also easy to maintain and breed in the laboratory, and hence, many inbred strains are now available for research. Studies on mice have contributed immeasurably to our understanding of cancer biology.
The development of transgenic, knockout, and knock-in mice has led to an exponential increase in their use as model organisms in research,...

Summary

Mice engineered to produce human interleukin-8 (hIL8) in intervertebral disc (IVD) tissues exhibited reduced activity and impaired natural behaviors, suggesting a role for hIL8 in pain-related functional decline.

Area of Science:

  • Biomedical Research
  • Molecular Biology
  • Animal Models

Background:

  • Interleukin-8 (IL8) is implicated in discogenic back pain.
  • Understanding the molecular mechanisms of IL8 signaling in pain is crucial.

Purpose of the Study:

  • To investigate the molecular mechanism of IL8 signaling in back pain.
  • To determine the effect of human IL8 (hIL8) expression on mouse activity and function.

Main Methods:

  • Generated a mouse model with conditional hIL8 expression in cartilage and intervertebral disc (IVD) tissues using GDF5-Cre.
  • Assessed functional capacities, including nest-making and natural behaviors.
  • Quantified immobility, eating duration, travel distance, and average speed.

Main Results:

  • hIL8-expressing mice showed significantly lower nesting scores compared to controls.
  • Mice expressing hIL8 spent more time immobile and eating during nighttime observation periods.
  • hIL8-expressing mice exhibited reduced travel distances and lower average speeds.

Conclusions:

  • Conditional expression of hIL8 in IVD tissues alters mouse behavior and reduces functional capacity.
  • This mouse model provides initial insights into the relationship between IL8 and pain-associated behavioral changes.
