Mesenchymal Stem Cells01:19

Mesenchymal Stem Cells

5.4K
Mesenchymal stem cells (MSCs) are adult stem cells that can differentiate into most connective tissue cell types, except for hematopoietic cells, depending upon the source of MSCs. For example, bone-marrow-derived MSCs (BM-MSCs) can differentiate into osteocytes, hepatocytes, and pancreatic and neuronal cells. MSCs can be isolated from various sources such as bone marrow, placenta, adipose tissue, teeth, and Wharton’s jelly, a gelatinous substance in the umbilical cord. The ease of their...

    Summary

Bone marrow mesenchymal stem cell (BM-MSC) soup enhances cell viability and growth. This BM-MSC soup also suppresses morphine-induced cell death in various cell lines.

    Area of Science:

    • Stem cell biology
    • Cellular communication
    • Drug-induced toxicity

    Background:

    • Bone marrow mesenchymal stem cells (BM-MSC) are recognized for therapeutic potential via secreted factors.
    • These factors mediate cellular communication, influencing cell differentiation and maturity.
    • Morphine can induce cell death, posing a challenge in therapeutic applications.

    Purpose of the Study:

    • To investigate the therapeutic potential of conditioned medium from BM-MSCs (mBM soup).
    • To determine the effect of mBM soup on morphine-induced cell death in PC12, U87, AGS, and MCF-7 cell lines.
    • To evaluate mBM soup's impact on cell viability, proliferation, and death pathways.

    Main Methods:

    • Harvesting conditioned media from BM-MSCs (mBM soup at 24h and 48h).
    • Assessing morphological changes using an inverted microscope.
    • Quantifying cell viability via trypan blue staining and MTT assay.
    • Determining cell death type using Hoescht / PI staining and analyzing NO secretion.

    Main Results:

    • mBM soup significantly increased cell viability in a time-dependent manner.
    • The conditioned medium demonstrated a protective effect against morphine-induced cell death.
    • NO secretion analysis and cell death assays provided insights into the underlying mechanisms.

    Conclusions:

    • mBM soup enhances the proliferation and growth of tested cell lines.
    • mBM soup effectively suppresses morphine-induced cell death.
    • These findings highlight the potential of BM-MSC-derived factors in mitigating drug toxicity.
