Altered IL-32 Signaling in Abdominal Aortic Aneurysm

Interleukin-32 (IL-32) is found in abdominal aortic aneurysm (AAA) tissue, particularly in smooth muscle cells and T-cells. This localized expression suggests IL-32 may play a role in AAA development.

Background:
- Interleukin-32 (IL-32) is a pro-inflammatory cytokine.
- Its role in abdominal aortic aneurysm (AAA) has not been previously investigated.
Purpose of the Study:
- To determine the expression and localization of IL-32 within human AAA tissue.
- To explore the potential involvement of IL-32 in the pathogenesis of AAA.
Main Methods:
- Immunohistochemistry and Western blot were used to analyze IL-32 protein expression in AAA and control aortic tissues.
- ELISA measured IL-32 levels in plasma from AAA patients and controls.
- Quantitative PCR (qPCR) assessed IL-32 mRNA expression in various cell types and aortic tissues.
Main Results:
- IL-32 protein and mRNA were predominantly expressed in smooth muscle cells (SMCs) and T-cell-rich areas within AAA tissues, especially in the intima/media layer.
- Expression was weaker in non-aneurysmal aortas.
- IL-32 expression was confirmed in isolated T cells, macrophages, endothelial cells, and SMCs, and could be induced by interferon-γ. Plasma IL-32 levels did not differ between AAA patients and controls.
Conclusions:
- IL-32 signaling is locally altered in abdominal aortic aneurysms.
- IL-32 may contribute to the development and progression of AAA.
- Further research, including animal models, is warranted to fully elucidate the role of IL-32 in AAA disease.
