  • To create novel reporter-expressing Zika virus (ZIKV) strains for antiviral screening.
  • To establish a molecular tool for identifying potential ZIKV therapeutics.

  • Engineered a full-length ZIKV cDNA clone in a bacterial artificial chromosome.
  • Inserted reporter genes (eGFP, NanoLuc luciferase) using an internal ribosome entry site.
  • Generated reporter-expressing ZIKV by transfecting BHK-21 cells with transcribed RNA.

  • Reporter-expressing ZIKVs exhibited slower growth and smaller foci than the parental virus.
  • Viral protein expression was largely identical, with novel larger forms of C and NS1 proteins observed.

Conclusions:

  • Reporter-expressing ZIKV strains are valuable tools for antiviral drug discovery.
  • These engineered viruses facilitate the screening and identification of potential ZIKV therapeutics.
