Mitochondrial pyruvate carrier (MPC) proteins, MPC1 and MPC2, are reduced in diabetic nephropathy (DN) patients. Lower MPC levels correlate with impaired kidney function in DN.
Area of Science:
- Nephrology
- Mitochondrial Biology
- Diabetology
Background:
- Mitochondrial dysfunction is implicated in diabetic nephropathy (DN) pathogenesis.
- Mitochondrial pyruvate carriers (MPC1 and MPC2) regulate pyruvate transport into mitochondria.
- Previous studies suggest potential benefits of increasing MPC in diabetic kidney disease models.
Purpose of the Study:
- To investigate the expression status of MPC1 and MPC2 in human patients with DN.
- To explore the correlation between MPC levels and clinical factors in DN patients.
Main Methods:
- Immunohistochemistry was used to examine MPC1 and MPC2 protein levels in renal tubules.
- Patients included primary glomerulonephropathy (PGN), PGN with diabetes mellitus (PGN-DM), and DN groups.
- Statistical analyses included Kruskal-Wallis test and Spearman's rank correlation.
Main Results:
- MPC1 and MPC2 were localized in renal tubules.
- DN patients exhibited significantly lower MPC1 and MPC2 levels compared to PGN and PGN-DM groups.
- MPC levels correlated significantly with serum creatinine, BUN, and eGFR in DN patients.
Conclusions:
- MPC proteins are decreased in the renal tubules of DN patients.
- Reduced MPC levels are associated with impaired renal function in DN.
- MPC expression may serve as a biomarker for DN progression.

Pyruvate Oxidation
First, the enzyme pyruvate dehydrogenase removes the carboxyl group from pyruvate and releases it as carbon dioxide. The stripped molecule is then oxidized and releases electrons, which are then picked up by NAD+...

Fates of Pyruvate
In aerobic organisms, pyruvate is metabolized via the citric acid cycle to produce reduced coenzymes NADH and FADH2. These coenzymes are then oxidized in the electron transport chain to produce ATP and, in the process, regenerate the NAD+ and FAD. As seen in some cell types and organisms, fermentation...
Allosteric Proteins-ATCase
Aspartate transcarbamoylase (ATCase) is a cytosolic enzyme that catalyzes the condensation of L-aspartate and carbamoyl phosphate to N-carbamoyl-L-aspartate. This reaction is the first step in pyrimidine biosynthesis. UTP and CTP, the end products of the pyrimidine synthesis...
