Ribosome Profiling02:24

Ribosome Profiling

4.0K
Ribosome profiling or ribo-sequencing is a deep sequencing technique that produces a snapshot of active translation in a cell. It selectively sequences the mRNAs protected by ribosomes to get an insight into a cell’s translation landscape at any given point in time.
Applications of ribosome profiling
Ribosome profiling has many applications, including in vivo monitoring of translation inside a particular organ or tissue type and quantifying new protein synthesis levels.
The technique...

Area of Science:

  • Genomics
  • Comparative transcriptomics
  • Animal biology

Background:

  • Gene differential expression studies are crucial for understanding animal life activities.
  • Previous giant panda research had limited transcriptome data, with only blood and spleen reported.

Purpose of the Study:

  • To explore transcriptome differences across various giant panda tissues.
  • To provide comprehensive transcriptome resources for the giant panda's heart, liver, lung, and kidney.

Main Methods:

  • RNA sequencing of heart, liver, spleen, lung, and kidney from five giant pandas.
  • Comparative analysis of intertissue gene expression patterns.
  • Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses, and protein-protein interaction (PPI) network construction.

Main Results:

  • Generated 194.52 GB of clean base data, identifying 18,701 genes, including 3,492 novel genes.
  • Identified 921 (heart), 553 (liver), 574 (spleen), 457 (lung), and 638 (kidney) tissue-specific differentially expressed genes (DEGs).
  • Found similar gene expression patterns between the spleen and lung, and identified key hub genes in each tissue.

Conclusions:

  • This study presents the first transcriptome data for the giant panda's heart, liver, lung, and kidney.
  • The findings offer valuable genetic resources for future research on giant pandas, aiding conservation efforts.
