A novel bacterial species, Pedobacter puniceum sp. nov. (HX-22-1^T), was identified from agricultural sludge. This Gram-negative bacterium expands the known diversity within the Pedobacter genus.
Area of Science:
- Microbiology
- Bacteriology
- Environmental Science
Background:
- The genus Pedobacter comprises Gram-negative bacteria found in various environments.
- Characterization of novel bacterial species is crucial for understanding microbial diversity and ecological roles.
Purpose of the Study:
- To isolate and characterize a novel bacterial strain from agricultural sludge.
- To determine the taxonomic position of the novel strain within the genus Pedobacter.
Main Methods:
- Isolation and cultivation of bacteria from sludge.
- Gram staining, morphology, and physiological tests.
- 16S rRNA gene sequencing and phylogenetic analysis.
- Genomic DNA G+C content determination.
- Chemotaxonomic analysis (fatty acids, ubiquinone, polar lipids).
- Genomic analysis (ANI and dDDH values).
Main Results:
- Strain HX-22-1^T is a Gram-negative, rod-shaped, non-spore-forming, non-flagellated bacterium.
- Phylogenetic analysis based on 16S rRNA gene sequences showed close relatedness to Pedobacter glucosidilyticus (98.63% similarity).
- Genomic analysis revealed ANI (89.6%) and dDDH (38.8%) values below the thresholds for species delineation.
- Optimal growth conditions were determined: 16-37°C, pH 6.0-8.0, and 0-1% NaCl.
- Chemotaxonomic data (fatty acids, ubiquinone, polar lipids) supported its placement within the genus Pedobacter.
Conclusions:
- Strain HX-22-1^T represents a novel species within the genus Pedobacter.
- The proposed name for this new species is Pedobacter puniceum sp. nov.
- The type strain is HX-22-1^T (KCTC 72655^T = CCTCC AB 2019348^T).
