Background:

  • Canine atopic dermatitis (cAD) is a common allergic skin disease in dogs, often caused by Dermatophagoides farinae.
  • Existing treatments for canine cAD frequently use allergen extracts developed for human immunotherapy, despite species-specific differences in sensitization.
  • There is a need for veterinary-specific immunotherapy enriched with major canine allergens.

Purpose of the Study:

  • To develop a specific allergen immunotherapy extract for veterinary use, enriched in canine major allergens from Dermatophagoides farinae.
  • To demonstrate the in vitro efficacy of this novel veterinary extract compared to a human extract.

Main Methods:

  • A veterinary Dermatophagoides farinae allergen extract was manufactured and characterized.
  • Protein profiles were analyzed using SDS-PAGE and size exclusion chromatography.
  • Major allergens Der f 15 and Der f 18 were quantified by mass spectrometry.
  • Allergenic profiles were assessed by immunoblot, and biological potency was determined using ELISA-inhibition assays.
  • The capacity of the extracts to induce cytokine production (IL-10, IFN-Ɣ) from canine peripheral blood mononuclear cells was evaluated.

Main Results:

  • The veterinary extract exhibited a higher concentration of high molecular weight proteins recognized by atopic dog sera.
  • Fold-increases in Der f 15 (2.07 ± 0.32) and Der f 18 (1.63 ± 0.15) were observed in the veterinary extract compared to the human extract.
  • The veterinary extract demonstrated significantly higher biological potency and induced greater production of IL-10 (1,780 pg/mL) and IFN-Ɣ (50.4 pg/mL) in vitro.

Conclusions:

  • A veterinary Dermatophagoides farinae extract specifically enriched with canine major allergens was successfully developed.
  • The developed veterinary extract demonstrated superior in vitro efficacy compared to a human extract, based on key immunological parameters.
  • This veterinary-specific extract holds promise for improved allergen immunotherapy in dogs with cAD.
