Summary

Circular RNAs (circRNAs) are globally downregulated in pediatric ependymomas. Differentially expressed circRNAs correlate with patient outcomes, suggesting their potential as diagnostic and prognostic biomarkers for these brain tumors.

Background:

  • Pediatric ependymomas, often in the cerebellum, lack specific therapies due to poorly understood pathobiology and few driver mutations.
  • Circular RNAs (circRNAs), crucial in tumorigenesis, have not been previously studied in pediatric ependymomas.

Discussion:

  • Next Generation Sequencing revealed global circRNA downregulation in ependymomas compared to controls.
  • Tumor suppressor circRNAs (circSMARCA5, circ-FBXW7) were significantly downregulated.
  • Hierarchical clustering separated patients based on prognosis, correlating with circRNA expression.

Key Insights:

  • CircRNA expression profiles differ between ependymomas and controls.
  • Distinct circRNA profiles were observed across pediatric brain tumor subtypes.
  • Specific circRNAs (from RMST, LRBA, WDR78, DRC1, BBS9) were upregulated in ependymomas.
