Insights

This retraction concerns a study on neural stem cell conditioned medium and its effect on Alzheimer's disease models. Data inconsistencies in figures and poor repeatability of results necessitate the retraction.

Area of Science:

  • Neuroscience
  • Cell Biology
  • Biochemistry

Background:

  • Alzheimer's disease is characterized by amyloid-beta (Aβ) peptide aggregation.
  • Neural stem cells (NSCs) show potential therapeutic properties.
  • Understanding molecular pathways involved in Aβ toxicity is crucial.

Purpose of the Study:

  • To investigate the protective effects of neural stem cell conditioned medium (NSCCM) against Aβ25-35 induced damage in SH-SY5Y cells.
  • To elucidate the role of the PCMT1/MST1 pathway in mediating these protective effects.

Main Methods:

  • SH-SY5Y cells were treated with Aβ25-35 to induce damage.
  • Cell viability and apoptosis assays were performed.
  • Western blotting was used to analyze protein expression, including PCMT1 and MST1.

Main Results:

  • The original study reported that NSCCM alleviated Aβ25-35 induced damage.
  • A proposed mechanism involved the upregulation of PCMT1 and activation of the MST1 pathway.
  • However, critical data errors were identified in Figure 1A and poor repeatability in PCMT1 expression data (Figure 2).

Conclusions:

  • Due to significant data inconsistencies and lack of repeatability, the findings of the original study cannot be validated.
  • The authors acknowledge the need to revise experimental designs and re-conduct experiments for verification.
  • This retraction highlights the importance of rigorous data integrity and reproducibility in scientific research.
