Pituitary adenylate cyclase-activating polypeptide (PACAP) may offer new therapeutic potential for age-related retinal conditions. This study found PACAP increased cell density in aging retinas, suggesting a protective effect against visual decline.
Area of Science:
- Neuroscience
- Ophthalmology
- Aging Research
Background:
- Retinal aging involves cellular damage and visual function decline.
- Somatostatin (SST) and PACAP are neuroprotective agents.
- A reliable transgenic model for SST-expressing amacrine cells is needed.
Purpose of the Study:
- Validate a transgenic model for SST-expressing amacrine cells.
- Investigate the chronic effect of PACAP on aging retinal SSTergic and dopaminergic cells.
Main Methods:
- Used SST-tdTomato transgenic mice aged 6, 12, and 18 months.
- Administered PACAP intravitreally every 3 months.
- Assessed SST and dopaminergic amacrine cell density in retinal whole mounts.
Main Results:
- PACAP treatment moderately elevated SST and dopaminergic cell densities in 12- and 18-month-old mice.
- Control retinas showed minor cell loss with age.
- Identified transgenic red fluorescence for SST-immunopositive amacrine cells.
Conclusions:
- A reliable transgenic model for studying SSTergic amacrine cells was developed.
- PACAP demonstrated potential to increase cell density in aged retinal tissue.
- PACAP may hold therapeutic promise for age-related retinal pathologies.
