Area of Science:

  • Veterinary Virology
  • Immunology
  • Swine Health

Background:

  • Porcine epidemic diarrhea virus (PEDV) causes severe disease in piglets, with current vaccines facing challenges due to viral variation and limited efficacy.
  • Existing PEDV vaccines have limitations including rapid viral mutation, insufficient immunity, and adverse effects, necessitating alternative prevention methods.

Purpose of the Study:

  • To develop and evaluate a novel antibody-based strategy for preventing PEDV infection in piglets.
  • To produce and characterize a monoclonal antibody targeting the PEDV S protein for potential therapeutic use.

Main Methods:

  • Production of an anti-PEDV S protein monoclonal antibody (anti-PEDV mAb-2) using hybridoma technology.
  • Testing the neutralizing activity of anti-PEDV mAb-2 against multiple PEDV strains (G1 and G2 genotypes).
  • In vivo challenge experiments in piglets to assess the efficacy of anti-PEDV mAb-2 in preventing PEDV infection.
  • Generation of HEK293 cell lines for the recombinant expression of anti-PEDV mAb-2.

Main Results:

  • The produced anti-PEDV mAb-2 demonstrated potent neutralization of PEDV strains from both G1 and G2 genotypes.
  • In vivo studies confirmed that anti-PEDV mAb-2 effectively inhibited PEDV infection in challenged piglets.
  • Stable HEK293 cell lines capable of expressing anti-PEDV mAb-2 were successfully established.

Conclusions:

  • Anti-PEDV mAb-2 derived from hybridoma supernatants provides effective protection against PEDV infection in piglets.
  • Recombinant expression of anti-PEDV mAb-2 in HEK293 cells offers a scalable platform for antibody production.
  • This antibody represents a promising candidate for enhancing PEDV prevention strategies in swine populations.
