Real Time RT-PCR

62.7K
Real-time reverse transcription-polymerase chain reaction, or Real-time RT-PCR, is an analytical tool used to determine the expression level of target genes. The method involves converting mRNA to complementary DNA with the help of an enzyme known as reverse transcriptase, followed by the PCR amplification of the cDNA. These two processes can be performed simultaneously in a single tube or separately as a two-step reaction.

Area of Science:

  • Molecular Biology
  • Genomics
  • Biochemistry

Background:

  • Long noncoding RNAs (lncRNAs) are RNA transcripts longer than 200 nucleotides with no protein-coding potential.
  • lncRNA expression is often tissue-specific and generally at lower levels compared to coding transcripts.
  • Accurate quantification of lncRNA expression is critical for functional characterization, particularly in differential expression studies.

Purpose of the Study:

  • To provide a detailed protocol for the sensitive and specific quantification of lncRNA expression.
  • To establish a reliable method for lncRNA quantitation using reverse transcription and SYBR Green real-time PCR.

Main Methods:

  • Reverse transcription of RNA to complementary DNA (cDNA).
  • Real-time quantitative PCR (qPCR) using SYBR Green dye chemistry.
  • SYBR Green dye binds to double-stranded DNA, emitting fluorescence proportional to amplicon quantity.

Main Results:

  • A detailed protocol for successful lncRNA quantitation is described.
  • The protocol leverages the simplicity and affordability of SYBR Green-based real-time PCR.
  • This method enables sensitive and specific measurement of lncRNA expression levels.

Conclusions:

  • The described protocol facilitates accurate lncRNA expression analysis.
  • This method supports functional studies of lncRNAs through precise quantification.
  • SYBR Green real-time PCR offers a cost-effective approach for lncRNA quantitation.
