Summary

Transurethral and suprapubic catheterization do not affect rat urethral function during leak point pressure testing. However, the first catheterization, regardless of method, temporarily increases external urethral sphincter electromyography activity.

Background:

  • Urethral function testing in rats often involves transurethral or suprapubic catheterization.
  • The impact of these methods and their order on urethral function remains unclear.

Purpose of the Study:

  • To compare the effects of transurethral versus suprapubic catheterization on leak point pressure (LPP) and external urethral sphincter electromyography (EUS EMG) in rats.
  • To determine if the order of catheterization influences LPP and EUS EMG results.

Main Methods:

  • A cross-over experimental design was employed in anesthetized female Sprague-Dawley rats.
  • Leak point pressure (LPP) and simultaneous external urethral sphincter electromyography (EUS EMG) were recorded.
  • Both transurethral and suprapubic catheterization methods were tested in a randomized order.

Main Results:

  • No significant differences in peak bladder pressure were observed between transurethral and suprapubic catheterization methods.
  • No significant differences in peak bladder pressure were found between the first and second catheter insertions.
  • Peak EUS EMG firing rate, amplitude, and amplitude difference were significantly higher after the first catheter insertion compared to the second, irrespective of the method used.

Conclusions:

  • The route of catheterization (transurethral or suprapubic) does not alter rat urethral function or create a functional partial outlet obstruction.
  • Either catheterization method is suitable for LPP and/or EUS EMG testing in rats.
  • Repeated catheterization may lead to transient changes in EUS EMG activity.
