Summary

Sleep disturbances are common in cognitive impairment, appearing even in early stages. Actigraphy can detect these sleep changes, suggesting potential for early intervention in Alzheimer's disease and mild cognitive impairment.

Background:

Purpose of the Study:

  • To investigate sleep characteristics using actigraphy in healthy controls, mild cognitive impairment (MCI), and mild-to-moderate AD patients.
  • To determine if sleep alterations are present in the early stages of cognitive decline.

Main Methods:

  • Actigraphic assessment of sleep parameters over 7 days.
  • Inclusion of 30 subjects: 10 controls, 10 MCI, and 10 mild-to-moderate AD.
  • Sleep quality assessed via Pittsburgh Sleep Quality Index (PSQI).

Main Results:

  • MCI patients showed increased sleep latency compared to controls (p=0.05).
  • AD patients exhibited significantly worse PSQI scores, longer time in bed, reduced total sleep time, increased sleep latency, decreased sleep efficiency, higher motor activity, and more wakefulness after sleep onset compared to controls.
  • Significant differences between AD and MCI groups were observed in sleep latency, wakefulness after sleep onset, sleep efficiency, number of awakenings, and motor activity.

Conclusions:

  • Sleep alterations are prevalent in cognitive impairment and are evident even in early stages.
  • Actigraphy provides clinically meaningful data for assessing sleep in cognitive decline.
  • Further research is warranted to explore if correcting sleep disturbances can impact cognitive decline progression.
