    Summary

    Autoimmune spondylodiscitis was induced in rats by immunizing them with nucleus pulposus and then injuring the tissue. This suggests nucleus pulposus can trigger an autoimmune response after injury.

    Background:

    • The intervertebral disc, specifically the nucleus pulposus, is normally immunologically privileged.
    • Understanding the mechanisms behind disc degeneration and inflammation is crucial for developing targeted therapies.

    Purpose of the Study:

    • To investigate the potential of nucleus pulposus to induce an autoimmune response leading to spondylodiscitis.
    • To determine the conditions under which nucleus pulposus can break immunologic tolerance.

    Main Methods:

    • Lewis rats were immunized with syngeneic nucleus pulposus in incomplete Freund's adjuvant.
    • In vivo injury of the nucleus pulposus was performed following immunization.
    • Control groups included rats subjected to injury alone or immunization alone.

    • Spondylodiscitis, characterized by mononuclear cell infiltration of vertebral discs, was observed in 100% of experimental animals following immunization and injury.
