A novel bacterium, Paracoccus shandongensis wg2T, was discovered in wastewater sludge. This aerobic, Gram-negative bacterium represents a new species within the Paracoccus genus, expanding microbial diversity knowledge.
Area of Science:
- Microbiology
- Bacteriology
- Environmental Science
Background:
- Wastewater treatment plants are crucial for environmental health.
- Microbial communities in activated sludge play vital roles in pollutant degradation.
- Characterizing novel bacteria aids in understanding microbial ecosystems and potential applications.
Purpose of the Study:
- To isolate and characterize a novel bacterial strain from activated sludge.
- To determine the taxonomic position of the novel strain using a polyphasic approach.
- To propose a new species and name for the identified bacterium.
Main Methods:
- Isolation and cultivation of bacteria from activated sludge.
- Physicochemical characterization (Gram staining, morphology, growth conditions).
- Phylogenetic analysis based on 16S rRNA gene sequencing.
- Genomic analysis (genome size, DNA G+C content).
- Chemotaxonomic analysis (respiratory quinones, fatty acids, polar lipids).
Main Results:
- A novel Gram-negative, aerobic, rod-shaped bacterium, strain wg2T, was isolated.
- Strain wg2T exhibited optimal growth at 30°C, 0-2.0% NaCl, and pH 7.0.
- Phylogenetic analysis placed strain wg2T within the genus Paracoccus, with high 16S rRNA gene similarity to P. sediminis and P. fontiphilus.
- Genomic and chemotaxonomic data supported its classification as a distinct species.
- The genome size was 3.93 Mbp with a DNA G+C content of 66.05%.
Conclusions:
- Strain wg2T represents a novel species of the genus Paracoccus.
- The proposed name for the novel species is *Paracoccus shandongensis* sp. nov.
- The type strain is wg2T (KCTC 72862T = CCTCC AB 2019401T).
