Confocal reflectance microscopy (CRM) offers a novel method for visualizing nanoparticles (NPs) in cells and tissues. This technique successfully tracked NP uptake and blood-brain barrier penetration, overcoming limitations of traditional imaging methods.
Area of Science:
- Biomedical Engineering
- Nanotechnology
- Microscopy
Background:
- Nanoparticle (NP) research requires effective visualization within biological systems.
- Current methods like electron microscopy and fluorescent tagging have significant limitations.
- There is a need for advanced imaging techniques for NP tracking.
Purpose of the Study:
- To evaluate Confocal Reflectance Microscopy (CRM) as an alternative imaging technique for NP visualization.
- To assess CRM's efficacy in tracking NP uptake and distribution in vitro and in vivo.
- To determine if CRM can differentiate NPs from non-specific signals.
Main Methods:
- Utilized CRM combined with fluorescence imaging.
- Visualized rhodamine B-conjugated silver NPs and FITC-conjugated lipid core-stearylamine NPs.
- Performed experiments both in vitro and in vivo.
Main Results:
- CRM successfully identified cellular uptake and blood-brain barrier penetration of NPs.
- NP-dependent reflectance signals showed dose and time dependency in vitro.
- CRM effectively distinguished specific NP signals from background fluorescence.
Conclusions:
- Confocal reflectance microscopy is a valuable tool for visualizing nanoparticles in biological tissues.
- CRM can be implemented using standard confocal microscopy equipment.
- This technique offers a promising alternative for NP imaging in research settings.
