Summary

Ribosomal RNA processing 12 homolog (RRP12) is upregulated in hepatocellular carcinoma, correlating with poor prognosis. Inhibiting RRP12 suppresses liver cancer cell proliferation and metastasis, suggesting RRP12 as a potential therapeutic target.

Area of Science:

  • Molecular Biology
  • Oncology
  • Genetics

Background:

  • Ribosomal RNA processing 12 homolog (RRP12) is a nucleolar protein essential for ribosome biogenesis.
  • RRP12 is implicated in the development of various cancers, but its role in hepatocellular carcinoma (HCC) remains unexplored.

Purpose of the Study:

  • To investigate the role and significance of RRP12 in hepatocellular carcinoma.
  • To determine RRP12's potential as a prognostic biomarker and therapeutic target in HCC.

Main Methods:

  • Pan-cancer analysis using TIMER and GEPIA databases.
  • Prognostic analysis and correlation with clinical parameters via GEPIA.
  • Investigation of RRP12 expression and function in liver cancer cell lines using Western Blot, scratch, Transwell, and Edu assays.

Main Results:

  • RRP12 was significantly upregulated across multiple cancers, including HCC.
  • RRP12 expression correlated with overall and disease-free survival rates in HCC patients.
  • RRP12 expression negatively correlated with tumor stage, grade, and size.
  • Knockdown of RRP12 inhibited proliferation, invasion, and metastasis of liver cancer cells.

Conclusions:

  • RRP12 plays a significant role in the occurrence and development of HCC.
  • RRP12 serves as a potential prognostic biomarker and therapeutic target for hepatocellular carcinoma.
