Complementation Tests00:49

Complementation Tests

5.4K
A complementation test is a simple cross to identify whether the two mutations are located on the same gene or different genes. It was first performed by Edward Lewis in the 1940s while working on fruit flies. He developed the test to identify the location and arrangement of different mutations on chromosomes.
Organisms heterozygous for different mutations are crossed pairwise in all combinations. If present on different genes, the mutations can complement each other by providing the missing...

Summary

Researchers identified a new gene, CYP71P1, responsible for disease lesion mimic (DLM) phenotypes in barley using a novel sequencing method. This discovery aids in understanding plant defense mechanisms and host-pathogen interactions.

Area of Science:

  • Plant genetics
  • Molecular biology
  • Crop science

Background:

  • Disease lesion mimic (DLM) mutants exhibit necrosis without pathogen infection, offering insights into plant defense pathways.
  • Traditional genetic strategies for identifying causal genes are time-consuming and resource-intensive.

Purpose of the Study:

  • To describe a collection of barley DLM mutants.
  • To introduce and validate a novel method, complementation by sequencing (CBS), for efficient gene identification in mutagenized populations.
  • To identify the gene responsible for specific DLM phenotypes in barley.

Main Methods:

  • Generation and characterization of 34 barley DLM mutants.
  • Application of complementation by sequencing (CBS) to identify causative genes.
  • Comparative phylogenetic analysis of identified gene families.

Main Results:

  • Successfully identified the cytochrome P450 CYP71P1 gene as responsible for orange blotch DLM mutants in barley.
  • Demonstrated that CBS is a cost-effective approach for cloning genes controlling phenotypes in chemically mutagenized collections.
  • Phylogenetic analysis revealed early emergence and recurrent loss of the CYP71P1 gene family in angiosperms.

Conclusions:

  • Complementation by sequencing (CBS) provides a powerful and efficient tool for gene discovery in plant research.
  • The identified CYP71P1 gene is crucial for understanding barley DLM phenotypes and potentially plant-pathogen interactions.
  • The TILLMore (TM) collection, combined with CBS, will advance research into DLM mechanisms and plant immunity.
