DICER-LIKE 1 (DCL1) and DICER-LIKE 4 (DCL4) prevent excessive 22-nucleotide small interfering RNAs (siRNAs) from endogenous genes. Loss of DCL1 and DCL4 leads to massive 22-nt genic siRNAs, disrupting plant development.
Area of Science:
- Plant molecular biology
- RNA interference pathways
- Gene regulation
Background:
- MicroRNAs (miRNAs) and small interfering RNAs (siRNAs) are key regulators of gene expression in plants.
- DICER-LIKE 2 (DCL2)-dependent 22-nt siRNAs are typically rare in wild-type Arabidopsis, with DCL1 and DCL4-dependent 21-nt RNAs being more abundant.
- The inhibitory role of DCL1 on endogenous 22-nt siRNA production remained unclear.
Purpose of the Study:
- To investigate the role of DCL1 in suppressing endogenous 22-nt siRNA production.
- To determine the impact of DCL1 and DCL4 on the abundance and characteristics of genic siRNAs.
- To identify the ARGONAUTE proteins involved in loading these siRNAs.
Main Methods:
- Analysis of small RNA populations in wild-type and dcl1 dcl4 double mutant Arabidopsis.
- Quantification of 22-nt genic siRNAs derived from specific genes (NIA1, NIA2, DGAT3, SMXL5, SMXL4).
- Identification of ARGONAUTE protein loading using immunoprecipitation assays.
Main Results:
- DCL1 and DCL2 were found to cleave miRNA precursors and dsRNAs from coding transcripts.
- A dcl1 dcl4 double mutant exhibited a significant increase (up to 95%) in 22-nt genic siRNAs from endogenous protein-coding genes.
- These abundant 22-nt genic siRNAs were primarily loaded into ARGONAUTE 1 (AGO1) or AGO2.
Conclusions:
- DCL1 and DCL4 play a crucial role in safeguarding post-transcriptional gene silencing.
- They prevent the overproduction of DCL2-dependent 22-nt genic siRNAs.
- Disruption of this balance by excessive 22-nt genic siRNAs can negatively impact plant growth and development.
