This study introduces a novel monoporous microsphere (MPM) carrier for delivering parathyroid hormone (PTH) to treat osteoporotic bone defects. The system precisely delivers PTH, reduces drug accumulation, and promotes bone regeneration while treating osteoporosis systemically.
Area of Science:
- Biomaterials Science
- Regenerative Medicine
- Drug Delivery Systems
Background:
- Osteoporotic bone defects require simultaneous treatment of osteoporosis and enhancement of bone regeneration.
- Conventional drug carriers often lead to suboptimal drug release and local accumulation, hindering effective treatment.
Purpose of the Study:
- To develop a novel monoporous microsphere (MPM) carrier for precise, in situ delivery of parathyroid hormone (PTH).
- To minimize topical drug accumulation and achieve systemic repair of osteoporotic bone defects.
Main Methods:
- Construction of specifically monoporous microspheres (MPMs) encapsulating dosage-sensitive, short half-time PTH.
- In vitro evaluation of drug release kinetics and carrier mobility.
- In vivo assessment of MPM-PTH efficacy in repairing cranial defects and treating systemic osteoporosis.
Main Results:
- MPMs demonstrated controlled, gradual release of PTH from their single pore, improving initial drug concentration.
- MPMs exhibited self-shifting behavior with animal movement, effectively reducing in vitro topical drug aggregation.
- In vivo studies confirmed that MPMs-PTH dual-acted to stimulate cranial defect regeneration and alleviate systemic osteoporosis.
Conclusions:
- A dynamically movable drug delivery system using monoporous microspheres was developed.
- This system enables precise in situ delivery, minimizes topical accumulation, and facilitates systematic repair of osteoporotic bone defects.

Calcium and Phosphorus
Calcium is a critical component of bones, especially in the form of calcium phosphate and calcium carbonate. Since the body cannot make calcium, it must be obtained from the diet. However, calcium cannot be absorbed from the small intestine without...
Bone Disorders
Bone deposition is also affected by the levels of sex hormones like estrogen and testosterone that promote osteoblast activity and bone matrix synthesis. When the level of these hormones decreases due to aging, it causes a reduction in bone deposition. As a result, bone resorption by osteoclasts...
