Real Time RT-PCR02:57

Real Time RT-PCR

58.6K
Real-time reverse transcription-polymerase chain reaction, or Real-time RT-PCR, is an analytical tool used to determine the expression level of target genes. The method involves converting mRNA to complementary DNA with the help of an enzyme known as reverse transcriptase, followed by the PCR amplification of the cDNA. These two processes can be performed simultaneously in a single tube or separately as a two-step reaction.
The real-time quantification of the number of amplified products is...

Area of Science:

  • Biotechnology
  • Molecular Biology
  • Analytical Chemistry

Background:

  • Polymerase Chain Reaction (PCR) is the gold standard for nucleic acid amplification.
  • Existing PCR instrumentation has limitations for point-of-need testing due to assay duration.
  • There is a need for rapid PCR systems to enable faster diagnostics.

Purpose of the Study:

  • To develop and present a novel instrument for ultra-fast real-time PCR.
  • To demonstrate rapid DNA and RNA amplification for point-of-need applications.
  • To reduce the time-to-result for PCR assays.

Main Methods:

  • A field-forward prototype instrument with custom microfluidic chips was designed.
  • A novel mechatronic system utilized two thermoelectric Peltiers for rapid heating and cooling (12 and 10 °C/s).
  • Synchronized thermal cycling and fluorescence monitoring were implemented.

Main Results:

  • The instrument completed 40 cycles of real-time PCR in under 10 minutes.
  • This represents an 84% reduction in time compared to standard 50-minute assays.
  • Robust amplification and detection of DNA and RNA targets were achieved.

Conclusions:

  • The developed rapid PCR system significantly accelerates nucleic acid amplification.
  • This technology enables faster, actionable information for diverse laboratory applications.
  • The system addresses the need for rapid, point-of-need testing solutions.
