This study developed a novel suspended self-nanoemulsifying drug delivery system (Su-SNEDDS) for alectinib hydrochloride (ALH). The Su-SNEDDS significantly enhanced ALH solubility and dissolution rates, offering a promising oral dosage form.
Area of Science:
- Pharmaceutical Technology
- Drug Delivery Systems
- Nanotechnology
Background:
- Alectinib hydrochloride (ALH) is a BCS class IV drug with poor water solubility.
- Enhancing ALH solubility and dissolution is crucial for effective oral delivery.
Purpose of the Study:
- To develop a novel suspended self-nanoemulsifying drug delivery system (Su-SNEDDS) for ALH.
- To improve the solubility and dissolution rate of ALH using Su-SNEDDS.
Main Methods:
- Preparation of Su-SNEDDS using Kolliphor HS 15 and Capmul MCM C8.
- Incorporation of micronized ALH particles (<5 μm) into the SNEDDS.
- Evaluation of dispersion, particle size, and dissolution rate in SLS buffer.
Main Results:
- The optimized SNEDDS showed complete dissolution within 30 min but low ALH loading (<0.6%).
- 10% ALH-loaded Su-SNEDDS achieved a 20-fold higher loading capacity.
- Su-SNEDDS demonstrated 100% dissolution rate within 30 min with spontaneous nanoemulsion formation.
Conclusions:
- Su-SNEDDS is an effective strategy to enhance ALH solubilization and dissolution.
- The developed system offers a novel technological approach for oral delivery of poorly soluble drugs.
- Micronized ALH particles within Su-SNEDDS readily disperse and solubilize.
