Adrenergic Receptors: β Subtype01:26

Adrenergic Receptors: β Subtype

1.9K
β-adrenoceptors have varied sensitivities towards adrenaline, noradrenaline, and isoprenaline. The order of agonist potency is as follows:
Isoprenaline > Adrenaline > Noradrenaline
Neurotransmitter binding to these receptors causes activation of adenylyl cyclase resulting in increased concentrations of cAMP and modulation of calcium ion channels within the cell. They are further classified into β1, β2, and β3 subtypes.

Adrenergic Antagonists: Pharmacological Actions of ɑ-Receptor Blockers

α-Adrenergic antagonists, known as α-blockers, exert their effects by inhibiting α-adrenoceptors, leading to specific physiological actions. α1-blockers and α2-blockers have distinct pharmacological actions and therapeutic applications.
α1-blockers: These drugs inhibit α1-adrenoceptors on smooth muscle cells, resulting in vasodilation. This vasodilation lowers blood pressure, making α1-blockers valuable in treating hypertension. Additionally,...

Adrenergic Antagonists: ɑ and β-Receptor Blockers

Third-generation β-blockers, such as labetalol and carvedilol, represent a significant advancement in managing cardiovascular conditions. Unlike conventional β-blockers, which can induce peripheral vasoconstriction, third-generation drugs block α1 adrenoceptors. This promotes vasodilation through several mechanisms, such as increased nitric oxide production, inhibition of calcium ion entry, opening of potassium ion channels, and antioxidant action. Labetalol, for instance, is...

Adrenergic Receptors: ɑ Subtype

1.7K
Adrenoceptors are classified into α and ꞵ classes based on their potencies to catecholamine agonists. α-adrenoceptors show the following order of catecholamine potency:
Adrenaline ≥ Noradrenaline >> Isoprenaline
α-adrenoceptors are further divided into α1 and α2-adrenoceptors.
α1-Adrenoceptors: These receptors are located postsynaptically on the effector organs and cause constriction of smooth muscle mediated by activation of phospholipase...

Adrenergic Receptors (Adrenoceptors): Classification

2.9K
Adrenergic receptors, or adrenoceptors, respond to the autonomic neurotransmitter noradrenaline and other endogenous catecholamine agonists. They are classified into two main families, α and β, based on their pharmacological response and are further subdivided depending on their location, elicited response, and affinity to specific agonists or antagonists.
α-Adrenoceptors
α-Adrenoceptors are classified into two main subtypes: α1 and α2. The α1 adrenoceptors,...

β-adrenergic antagonists, or β-blockers, modulate the sympathetic nervous system by targeting β-adrenoceptors and inhibiting catecholamine-mediated sympathetic responses. β-blockers differ in their adrenoceptor subtype affinity, lipophilicity, and α-blocking capabilities. The history of β-blocker development began with the prototype, dichloroisoprenaline, which exhibited partial agonist activity. As a result, propranolol was developed as a pure antagonist but...

Summary

The common β3-adrenoceptor (AR) 64Arg variant may be linked to overactive bladder (OAB), but evidence is inconsistent. Further research is needed to confirm if this polymorphism affects β3-AR function and OAB treatment response.

Area of Science:

  • Pharmacogenomics
  • Urology
  • Molecular Biology

Background:

  • Beta-3 adrenoceptors (ARs) are a key drug target for overactive bladder (OAB).
  • The human β3-AR gene exhibits common polymorphisms, notably the tryptophan (Trp) to arginine (Arg) exchange at position 64 (64Arg).

Purpose of the Study:

  • To review the impact of β3-AR polymorphisms on urological diseases, particularly OAB.
  • To evaluate the association between the β3-AR 64Arg variant and OAB prevalence and symptom severity.

Main Methods:

  • Narrative review of existing literature.
  • Analysis of studies investigating the 64Arg allele frequency in OAB patients versus controls.
  • Examination of in vitro and in vivo studies on receptor function and agonist response.

Main Results:

  • Inconsistent findings regarding the association between the 64Arg allele and OAB prevalence.
  • Limited differences in symptom severity between 64Arg and 64Trp carriers.
  • In vitro studies suggest potential hypofunctionality of the 64Arg variant, but results are not always confirmed.

Conclusions:

  • Evidence suggests carriers of the 64Arg genotype may have fewer or hypofunctional β3-ARs, potentially linked to OAB.
  • Observed associations are inconsistent, possibly due to insufficient statistical power.
  • Larger, well-powered studies are required to definitively assess the functional impact of the 64Arg variant on β3-AR activity and OAB.
