Targeting the connexin 43 (Cx43) protein may prevent or reduce post-surgical adhesions. Cx43 upregulation correlated with adhesion severity, and its inhibition decreased adhesion formation in a mouse model.
Area of Science:
- Biomedical Science
- Cell Biology
- Surgical Innovation
Background:
- Post-surgical peritoneal adhesions significantly impair quality of life and fertility.
- Current methods for preventing adhesions are ineffective.
- Connexin 43 (Cx43), a gap junction protein, is implicated in fibrosis and inflammation-related conditions.
Purpose of the Study:
- To investigate the dynamic expression of Cx43 in a mouse model of surgical adhesions.
- To evaluate the therapeutic potential of targeting Cx43 to prevent adhesion formation.
Main Methods:
- Utilized a mouse ischemic button model to induce surgical adhesions.
- Administered Cx43 antisense via Pluronic gel to reduce Cx43 expression.
- Assessed adhesion severity, Cx43 and TGF-β1 levels via immunofluorescence, and tissue plasminogen activator concentration via ELISA.
Main Results:
- Cx43 levels significantly increased within hours of adhesion formation, correlating with increasing adhesion severity.
- Cx43 upregulation was observed alongside robust, opaque, and vascularized adhesions by 7 days.
- Cx43 antisense treatment attenuated Cx43 upregulation and reduced the number and severity of adhesions.
Conclusions:
- Cx43 expression dynamically increases during surgical adhesion development.
- Inhibiting Cx43 expression shows promise in preventing or reducing the severity of post-surgical adhesions.
- Targeting Cx43 represents a potential therapeutic strategy for mitigating adhesion-related complications.
