Ribosome Profiling02:24

Ribosome Profiling

3.6K
Ribosome profiling or ribo-sequencing is a deep sequencing technique that produces a snapshot of active translation in a cell. It selectively sequences the mRNAs protected by ribosomes to get an insight into a cell’s translation landscape at any given point in time.
Applications of ribosome profiling
Ribosome profiling has many applications, including in vivo monitoring of translation inside a particular organ or tissue type and quantifying new protein synthesis levels.

RNA Splicing01:32

RNA Splicing

56.6K
Splicing is the process by which eukaryotic RNA is edited before its translation into protein. The RNA strand transcribed from eukaryotic DNA is called the primary transcript. The primary transcripts that become mRNAs are called precursor messenger RNAs (pre-mRNAs). Eukaryotic pre-mRNA contains alternating sequences of exons and introns. Exons are nucleotide sequences that code for proteins, whereas introns are the non-coding regions. In RNA splicing, introns are removed and exons are bonded...

Area of Science:

  • Molecular Biology
  • Genetics
  • Oncology

Background:

  • Long noncoding RNAs (lncRNAs) are crucial in disease development, including cancer.
  • Intronic transcripts, a type of lncRNA within protein-coding genes, can encode micropeptides.
  • Limited research exists on intronic transcript expression in osteosarcoma.

Purpose of the Study:

  • To investigate differentially expressed intronic transcripts between osteosarcoma and normal bone tissues.
  • To identify potential micropeptide-encoding intronic transcripts in osteosarcoma.
  • To determine the cellular localization of these intronic transcripts.

Main Methods:

  • Comparative analysis of intronic transcript expression profiles.
  • Identification of differentially expressed genes using transcriptomic data.
  • Bioinformatic analysis for micropeptide encoding potential and subcellular localization.

Main Results:

  • NRG1-IT1, FGF14-IT1, and HAO2-IT1 were downregulated in osteosarcoma.
  • ER3-IT1, SND1-IT1, ANKRD44-IT1, AGAP1-IT1, DIP2A-IT1, LMO7DN-IT1, SLIT2-IT1, RNF216-IT1, and TCF7L1-IT1 were upregulated in osteosarcoma.
  • The micropeptide encoding potential and cellular locations of these transcripts were identified.

Conclusions:

  • Specific intronic transcripts show altered expression patterns in osteosarcoma.
  • These findings contribute to understanding the role of lncRNAs in osteosarcoma.
  • Identified intronic transcripts may serve as potential diagnostic or therapeutic targets for osteosarcoma.
