Purpose of the Study:

  • To investigate the causal relationship between systemic iron status and the risk of 24 specific mental disorders using Mendelian randomization.
  • To differentiate potential causal effects from observational biases.

Main Methods:

  • A two-sample Mendelian randomization (MR) analysis was employed.
  • Genetic variants associated with four iron biomarkers (serum iron, ferritin, transferrin saturation, transferrin) were utilized.
  • Summary-level data for mental disorders were sourced from the UK Biobank, with inverse-variance weighted (IVW) and sensitivity analyses performed.

Main Results:

  • Genetically predicted higher serum iron, ferritin, and transferrin saturation showed positive associations with depression and psychogenic disorder.
  • Conversely, higher iron status markers were inversely associated with gender identity disorders.
  • Increased transferrin (indicating lower iron status) was linked to higher risk of gender identity disorders and lower risk of psychogenic disorder.

Conclusions:

  • Higher iron status appears to have a detrimental causal effect on depression and psychogenic disorder.
  • Elevated iron status may offer a protective effect against the risk of gender identity disorders.
  • Further research into the underlying biological mechanisms is recommended to inform preventative strategies for mental health.
