This study identifies key genes (HMOX1, LOX, ACTG1, STAT3, GNB5) linked to pulpitis using weighted gene co-expression network analysis (WGCNA). Dental pulp capping therapy demonstrated potential to reverse the expression of some identified genes.
Area of Science:
- Biomedical research
- Genomics
- Systems biology
Background:
- Pulpitis, a common bacterial dental disease, presents diagnostic challenges with conventional methods relying on subjective clinical symptoms.
- Accurate and rapid diagnosis of pulpitis is crucial for effective therapeutic decisions.
Purpose of the Study:
- To identify key genes associated with pulpitis through integrated microarray data analysis and systems biology approaches.
- To apply weighted gene co-expression network analysis (WGCNA) for uncovering molecular mechanisms of pulpitis.
Main Methods:
- Microarray data from inflamed and normal dental pulp samples were analyzed using WGCNA to construct a gene co-expression network.
- Hub genes within the module most correlated with pulpitis were identified.
- Validation of identified hub genes was performed using reverse transcription-quantitative polymerase chain reaction (RT-qPCR) in a rat pulpitis model with pulp capping.
Main Results:
- WGCNA identified 22 gene modules, with the 'darkgrey' module showing the highest correlation to pulpitis.
- Five hub genes (HMOX1, LOX, ACTG1, STAT3, GNB5) were identified as significantly associated with pulpitis.
- RT-qPCR confirmed differential expression of these hub genes in inflamed pulp, and pulp capping partially reversed HMOX1, LOX, and ACTG1 expression.
Conclusions:
- This study provides a comprehensive view of pulpitis by identifying and validating key genes using WGCNA.
- Pulp capping with iRoot BP plus shows potential to modulate the expression of specific genes involved in pulpitis.
