Area of Science:

  • Microscopy
  • Bioimage Analysis
  • Computational Biology

Background:

  • Sample drift in 3D microscopy hinders accurate temporal analysis of biological processes.
  • Existing open-source solutions for 3D drift correction are often slow and complex.

Purpose of the Study:

  • To introduce Fast4DReg, a novel, rapid, and open-source tool for correcting axial and lateral drift in 3D video-microscopy datasets.
  • To provide a user-friendly solution for improving the quality of 3D imaging data.

Main Methods:

  • Fast4DReg utilizes intensity projections along multiple axes.
  • It estimates frame-to-frame drift using 2D cross-correlations.
  • The tool can also register misaligned channels in 3D datasets.

Main Results:

  • Fast4DReg demonstrates superior performance and speed compared to existing state-of-the-art open-source drift-correction tools.
  • The tool effectively corrects both sample drift and channel misalignment in 3D microscopy data.
  • Validation was performed using both synthetic and acquired datasets.

Conclusions:

  • Fast4DReg offers a significant advancement in processing 3D microscopy videos.
  • The tool provides a fast, easy-to-use method for correcting drift and improving 3D image data quality for subsequent analysis.
  • This enhances the reliability of temporal visualization and quantification in microscopy experiments.
