Background:

  • Existing resting-state fMRI (rs-fMRI) studies show inconsistencies due to small sample sizes and lack of follow-up data.

Purpose of the Study:

  • To investigate functional impairments and neural reorganization in the language network of PSA patients.
  • To provide robust evidence using a moderate sample size and follow-up data.

Main Methods:

  • Utilized rs-fMRI to assess Amplitude of Low Frequency Fluctuations (ALFF) and functional connectivity.
  • Included 35 PSA patients and 35 healthy controls at baseline, with 16 patients followed up.
  • Analyzed alterations in the left inferior frontal gyrus (IFG), supplementary motor area (SMA), and superior temporal gyrus (STG).

Main Results:

  • Baseline PSA patients exhibited decreased ALFF in the left IFG and reduced IFG connectivity with bilateral SMA and right STG compared to controls.
  • Follow-up data showed enhancement in decreased ALFF and IFG connectivity among PSA patients.
  • These neural improvements were synchronized with language performance recovery.

Conclusions:

  • PSA is characterized by reduced intrinsic neural activity and inter-connections within the language network.
  • Neural activity and connectivity normalize as language performance improves, suggesting effective brain plasticity.
