Selectins01:25

Selectins

3.4K
Cell adhesion is  an essential aspect of multicellularity. While stable cell interactions usually occur between cells of the same type, transient cell interactions occur between cells of different tissue types, such as between neutrophils and endothelial cells. Selectins are one class of cell adhesion molecules (CAMs) that bind carbohydrate ligands to form transient cell adhesion. They are rod-like proteins with a long extracellular part of variable length ending with the lectin domain,...

Protein Networks02:26

Protein Networks

4.0K
An organism can have thousands of different proteins, and these proteins must cooperate to ensure the health of an organism. Proteins bind to other proteins and form complexes to carry out their functions. Many proteins interact with multiple other proteins creating a complex network of protein interactions.
These interactions can be represented through maps depicting protein-protein interaction networks, represented as nodes and edges. Nodes are circles that are representative of a protein,...

Targeted Cancer Therapies

7.7K
The targeted cancer therapies, also known as “molecular targeted therapies,” take advantage of the molecular and genetic differences between the cancer cells and the normal cells. It needs a thorough understanding of the cancer cells to develop drugs that can target specific molecular aspects that drive the growth, progression, and spread of cancer cells without affecting the growth and survival of other normal cells in the body.
There are several types of targeted therapies against...

Cancer-Critical Genes II: Tumor Suppressor Genes

7.6K
Genes usually encode proteins necessary for the proper functioning of a healthy cell. Mutations can often cause changes to the gene expression pattern, thereby altering the phenotype.
When the function of certain critical genes, especially those involved in cell cycle regulation and cell growth signaling cascades, gets disrupted, it upsets the cell cycle progression. Such cells with unchecked cell cycles start proliferating uncontrollably and eventually develop into tumors.
Such genes that act...

Cancer Stem Cells and Tumor Maintenance02:40

Cancer Stem Cells and Tumor Maintenance

5.0K
Early diagnosis and treatment can often cure cancer. However, even with treatment, residual cells called cancer stem cells (CSC) might remain, often causing tumor recurrence. These cancer stem cells possess the potential for self-renewal and multi-lineage differentiation and are often responsible for the therapeutic resistance displayed in most cancers.
Cancer stem cells are thought to originate from tissue-specific normal stem cells or progenitor cells. The normal stem cells usually reside in...

Single-Strand DNA Binding Proteins01:03

Single-Strand DNA Binding Proteins

14.5K
For successful DNA replication, the unwinding of double-stranded DNA must be accompanied by stabilization and protection of the separated single strands of the DNA. This crucial task is performed by single-strand DNA-binding (SSB) proteins. They bind to the DNA in a sequence-independent manner, which means that the nitrogenous bases of the DNA need not be present in a specific order for binding of SSB proteins to it. The binding of SSB proteins straightens single-stranded DNA (ssDNA) and makes...

Summary

Specificity proteins (Sp) 1, 3, and 4 are crucial for cancer development and growth. Targeting these transcription factors, even individually, shows promise for new cancer therapies.

Area of Science:

  • Oncology
  • Molecular Biology
  • Gene Regulation

Background:

  • Specificity proteins (Sp) 1, 2, 3, and 4 share similarities in cancer cells.
  • Sp1 is a negative prognostic factor in numerous tumor types.
  • Sp transcription factors (TFs) regulate pro-oncogenic factors and pathways.

Purpose of the Study:

  • Review the roles of Sp1, Sp3, and Sp4 in cancer development.
  • Discuss their regulation of oncogenic pathways and interactions with non-coding RNAs.
  • Examine the potential of targeting Sp TFs for cancer therapy.

Main Methods:

  • Reviewed studies on normal cell transformation into cancer cell lines.
  • Analyzed knockdown studies of Sp TFs in cancer cell lines.
  • Investigated Sp TF interactions with non-coding RNAs (microRNAs and long non-coding RNAs).

Main Results:

  • Increased Sp1 levels accompany normal cell transformation into cancer.
  • Silencing individual Sp1, Sp3, or Sp4 TFs reduced cancer growth, invasion, and induced apoptosis.
  • Sp TFs are non-oncogene addicted, as silencing one is not compensated by others.
  • Sp1 interacts with non-coding RNAs to promote oncogenic functions.

Conclusions:

  • Sp1, Sp3, and Sp4 play significant roles in cancer progression.
  • These Sp TFs are non-oncogene addicted, making them potential therapeutic targets.
  • Agents targeting Sp TFs, especially in combination therapies, warrant further clinical investigation for enhanced efficacy and reduced toxicity.
