Summary

Hepatitis B virus-related liver cancer (HBV-related HCC) shows decreased 5-hydroxymethylcytosine (5-hmC) levels. This epigenetic change is linked to reduced TET1 and IDH2 expression in tumor tissues.

Background:

  • Decreased 5-hydroxymethylcytosine (5-hmC) is observed in various cancers.
  • Limited data exists on 5-hmC levels in hepatitis B virus-related hepatocellular carcinoma (HBV-related HCC).

Purpose of the Study:

  • To investigate 5-hmC levels in HBV-related HCC tumor tissues.
  • To explore the underlying mechanisms, including TET and IDH enzyme expression, contributing to 5-hmC changes in HBV-related HCC.

Main Methods:

  • Immunohistochemistry was used to quantify 5-hmC, 5-methylcytosine, TET1, and IDH2 expression.
  • Image Pro Plus 6.0 software analyzed protein expression in tumor and adjacent para-cancerous tissues from 40 HBV-related HCC patients.

Main Results:

  • A significant decrease in 5-hmC and an increase in 5-methylcytosine were observed in HBV-related HCC tumor tissues.
  • Expression levels of TET1 and IDH2 were reduced in tumor tissues.
  • The reduction in TET1 and IDH2 showed a positive correlation with decreased 5-hmC levels.

Conclusions:

  • Reduced 5-hmC is an epigenetic hallmark of HBV-related HCC.
  • The deficiency of 5-hmC in HBV-related HCC is primarily attributed to the decreased expression of TET1 and IDH2.
