Purpose of the Study:

  • To isolate and identify lignin-degrading bacteria from paper mill sludge.
  • To assess the laccase activity of isolated bacterial species.

Main Methods:

  • Isolation of bacteria from paper mill sludge using Kraft lignin as the sole carbon source.
  • Determination of laccase activity via oxidation of 2,2'-azinobis-(3-etilbencenotiazolina-6-sulfonate) (ABTS).
  • Identification of bacterial species using molecular biology techniques.

Main Results:

  • Seven bacterial species with lignin-degrading and laccase-producing capabilities were identified.
  • Agrobacterium tumefasciens, Klebsiella grimontii, and Beijeinckia fluminensis were identified, with the latter two reported for the first time.
  • K. grimontii and B. fluminensis exhibited the highest laccase activities (0.319 ± 0.005 UmL-1 and 0.329 ± 0.004 UmL-1, respectively).

Conclusions:

  • Paper mill sludge is a viable source for isolating lignin-degrading bacteria with significant laccase activity.
  • The identified bacteria hold potential for biotechnological applications, particularly in lignin valorization and bioremediation.
