    Immune complexes (ICs) trigger acute synovitis in rabbit knees, activating polymorphonuclear granulocytes (PMNGs) within joints. Activated PMNGs show degranulation and phagocytosis, indicating a two-phase leukocyte response in joint inflammation.

    Background:

    • Acute synovitis is an inflammatory joint condition.

    Purpose of the Study:

    • To investigate the behavior and activation of leukocytes during IC-induced acute synovitis in rabbit knee joints.
    • To elucidate the phases of leukocyte response in IC-induced joint inflammation.

    Main Methods:

    • Induction of acute synovitis via intraarticular injection of bovine serum albumin (BSA)-anti BSA immune complexes (ICs) in rabbits.
    • Morphological analysis (electron microscopy) and chemiluminescence (CL) measurements of emigrated leukocytes.
    • In vitro stimulation assays and inhibition experiments to assess leukocyte responsiveness and metabolic pathways.

    Main Results:

    • Leukocytes migrating into IC-injected joints showed degranulation and IC phagocytosis, indicating activation.
    • These activated leukocytes exhibited reduced CL response to in vitro IC stimulation compared to other leukocyte sources.
    • Oxygen radicals and lipoxygenase pathway metabolites were involved in the CL response of IC-stimulated joint fluid leukocytes.

    Conclusions:

    • Leukocyte reaction in IC-induced synovitis involves an initial migration phase without increased CL.
    • A subsequent activation phase is characterized by degranulation, IC phagocytosis, and increased CL.
    • Leukocyte responsiveness is modified in a stimulus-specific manner during IC-induced joint inflammation.
