    Area of Science:

    • Food Science
    • Analytical Chemistry
    • Biochemistry

    Background:

    • Alkaline phosphatase is an enzyme found in casein.
    • Accurate determination of residual alkaline phosphatase is crucial for casein quality control.
    • Existing sample preparation methods may be time-consuming.

    Purpose of the Study:

    • To compare two distinct sample preparation techniques for alkaline phosphatase determination in casein.
    • To validate a rapid colorimetric test for residual phosphatase analysis.
    • To assess the efficiency and accuracy of a new rapid sample preparation method.

    Main Methods:

    • A collaborative study involving seven laboratories.
    • Testing of ten unknown casein products with varying residual phosphatase levels.

    Related Experiment Videos

  • Application of two different sample preparation techniques followed by a rapid colorimetric assay.
  • Main Results:

    • No significant difference in phosphatase content was observed between the two preparation methods.
    • 100% accuracy in analyzing ground casein samples.
    • 98% accuracy in analyzing unground casein samples.

    Conclusions:

    • The alternative rapid sample preparation method is effective and accurate for alkaline phosphatase determination in casein.
    • The rapid method provides reliable results comparable to traditional techniques.
    • Official adoption of the rapid sample preparation method facilitates faster quality control processes.
